Quantitative evaluation of 31P MRS of Tissue Samples using Magic Angle Spinning

High-resolution 31P MAS has been compared with high-resolution MRS of extracts of the same tissue samples. MAS produces spectra of superior signal-to-noise ratio, with concentration estimates in good agreement with those of tissue extracts, but leaving tissue intact for histological analysis. Concentration estimates using reference compound acquired in a separate measurement are more reliable than adding compound to tissue within the rotor. The method is well suited to the study of tumour biopsies where only small tissue samples are available and where histological verification is required, for example before and after treatment with new targeted agents that affect phospholipid metabolism
