Metabolites and Macromolecules in healthy aging and subjective cognitive decline. A longitudinal 7 T 1H-MRS study

Synopsis

Motivation: The literature on concentrations of human brain metabolites and macromolecules (MM) during healthy aging is inconsistent. Studies on the neurochemical development in aging individuals with Subjective Cognitive Decline (SCD) are also rare. No study has investigated macromolecule levels in SCD.

Goal(s): Differentiate neurochemical changes in metabolites and MM in healthy aging and SCD using MRS.

Approach: Longitudinal 7T-MRS data, parametric MM model, and weighted linear mixed model

Results: Significant changes in GABA and specific MM peaks in SCD, suggesting a link to Alzheimer Disease. No significant aging effect on metabolites, though moderate increase for tCho and tCr. Influence of sex on metabolite levels

Impact: Our results highlight the potential of macromolecules as biomarkers, also pointing to a biochemical link between SCD and AD.
