1 H-[ 13 C]-NMR Studies of Cerebral Metabolism in Chronic Unpredictable Mild Stress Model of Depression

The molecular mechanism responsible for depression is still elusive. The present study evaluate neural metabolism in the prefrontal cortex of mice subjected to chronic unpredictable mild stress (CUMS). Sucrose preference and forced swim test were carried out to assess the behavioral phenotype. The neuronal and astroglial metabolism were monitored by 1 H-[ 13 C]-NMR spectroscopy together with infusion of [1,6- 13 C 2 ]glucose and [2- 13 C]acetate, respectively. Mice subjected to CUMS exhibit increased immobility in the forced swim test and less sucrose preference which are typical phenotype of depression. Most importantly, the glutamatergic, GABAergic and astroglial metabolic activities were found to decreased in CUMS treated mice.
